Introduction

Use of Natural Killer cells for treatment of leukemia, infections or autoimmune diseases.

Factors influencing NK cell activity

Poggi et al (2005 [1]) reported that coculture of IL-2 activated NK cells and the tumor cell line Fo1 resulted in NCR mediated induction of apoptosis at low effector to target ratios.. Addition of Cyclosphorin A (optimal concentration 500 ng/ml) to in vitro cultures reduced sFasL release and blocked induction of apoptosis whithout affecting the triggering of NK cell mediated cytotoxicity towards tumor cells. This effect might propose the use of CsA in combination with low amounts of NK cells for immunotherapy avoiding their limited maintenance due to tumor cell induced NK cell apoptosis.
