Introduction
NKG2D is one of the major triggering receptors of NK cells. Garrity et al. (2005) demonstrated that one NKG2D homodimer assembles with four DAP10 chains which may result in triggering also at low ligand density on target cells as binding of a single ligand can result in phosphorylation of four DAP10 chains.

Ligands
Known NKG2D-ligands in humans are MICA, MICB, ULBP1, ULBP2, ULBP3, and more recently MICC and ULBP4.
