KIR phenotyping of human CD56+CD3+ NKT cells Ilka Bondzio, Andreas Arendt, Jürgen Schmitz, Volker Huppert

Conclusion: Human CD3+CD56+ NKT cells (both unseparated and separated using MACS® Technology) are positive for several KIRs but diff er from NK cells in the frequencies of KIR+ subsets. Further research will be required to explore these diff erences as they may point to diff erent mechanisms of KIR regulation. The identifi cation of KIR-positive NKT cells may also provide an opportunity for their use for functional KIR studies instead of NK cell clones, as the cloning of NKT cells may prove easier (i.e. using standard T cell cloning methods) than that of NK cells. Further KIR phenotyping of KIR-selected NKT cell clones can also be performed on the mRNA level using the KIR Typing Kit (Miltenyi Biotec). This kit would also be invaluable for the evaluation of anti- KIR antibodies with unknown specificities.
