Introduction

Killer cell immunoglobulin-like receptors (KIR) are polymorphic receptors on human NK cells and a subset of T cells for MHC class I molecules and additional but yet unknown ligands. They are responsible for the detection of "missing self" of virus infected and transformed cells by NK cells by surveilling the level of MHC class I molecules. Functionally similar but structurally totally different molecules in mice belong to the Ly49 family. KIRs can be both inhibitory (long cytoplasmatic chain: KIR2DL, KIR3DL) and activating (short cytoplasmatic chain: KIR2DS, KIR3DS). The unique KIR2DL4 molecule can have both inhibitory as well as activating function. KIR Haplotypes can be divided into group A and group B ones. Group A KIR haplotypes have only one activating KIR (i.e. KIR2DS4), Group B KIR haplotypes can have up to five (of six) activating KIRs (Hsu et al., J Immunol 2002; Maxwell et al., Tissue Antigens 2002; Abi-Rached et al., J Exp Med 2005)

KIR History

The activating KIRs have evolved from the ancestral inhibitory KIRs, as have similar receptors (Ly49) in rodents. Upon this single event, additional activating receptors have been generated through recombination and gene duplication. (Abi-Rached et al., J Exp Med 2005)

KIR in rodents and Ly49 in humans

The human genome contains a single Ly49 gene, which is nonfunctional (Abi-Rached et al., J Exp Med 2005). Rodent genomes contain KIR genes, which seem to be functionally different from the hoinoid KIR gene family (Welch et al., 2003; Abi-Rached et al., J Exp Med 2005).

Heider, Cremer et al. (2005, poster presentation NK Hawaii [3]) described a novel commercial kit that can be used for both KIR Genotyping and KIR mRNA expression profiling due to a specific primer design.
