Introduction

Gamma/delta T cells are part of the innate immune system. They exhibit a MHC-unrestricted lytic activity against different tumor cells in vitro (Wilhelm et al., 2003; Kunzmann et al., 2000; Sicard et al., 2001; Kato et al., 2001) similar to NK cells. Gamma/delta T cells can be activated by synthetic aminobisphosphonates (e.g. Novartis' Pamidronate and can be expanded both in vitro and in vivo by a combination of Pamidronate and low dose IL-2 (Wilhelm et al., 2003)

gamma/delta T cells and diseases

In vivo activation and expansion of gamma/delta T cells has been used by Wilhelm et al. (Blood 2003) to treat NHL or MM patients previously tested for in vitro proliferation of gamma/delta T cells upon addition of bisphosphonate and IL-2. In vivo proliferation was observed in 5 of 9 patients (pre-screened for in vitro proliferation of gamma/delta T cells) and clinical responses were observed in 3 of these 5 patients.

They demonstrated that fewer patients with hematologic malignancies than a control group of healthty volunteers (49% vs. 88%) showed pamidronate/IL-2 induced gamma/delta T cell proliferation in vitro, which might be due to prior chemotherapy in these patients.

Kunzmann and Wilhelm (2005, [1]) review evidence of pre-clinical and early clinical studies regarding the contribution of gamma/delta T cells in the defense against lymphoid malignancies. They describe that Vd2Vg9 T cells exert cytotoxic activity against lymphoid malignancies, pointing to potential use of stem cell grafts that contain gamma/delta T cells or gamma/delta T cell donor lymphocyte infusions.
