Regulatory NK cell subset, mainly producing cytokines rather than killing of virus infected or cancer cells.

CD56bright NK cells and diseases

Activated (CD69+) CD56bright NK cells may play a role in liver inflammation in hepatitis C virus infected patients (Lin et al., 2004)

Katchar et al. (2005 [1]) found that the majority of lung NK cells in patients with pulmonary sarcoidosis are CD56bright. Most lung NK cells expressed HLA-E-specific inhibitory receptors (i.e. CD94/NKG2A), but only a few killer cell immunoglobulin-like inhibitory receptors specific for HLA-A/B/C.

Gene expression profiling CD56bright versus CD56 dim NK cells

Koopman et al. (2003, [2]) compared gene expression of decidual versus peripheral blood NK cells and included gene expression data of CD56bright versus CD56dim NK cell subsets. Genes are listed in the order of x-fold change of bright vs. dim cells. Significant differences are listed in bold.

  • higher expression in bright cells
    • RANK (12 fold), BMP2, CD9, IFITM3, HLA-DRA, ITGAX, CD44, KIAA1775, IL7R, CD2-AP, SPRY 2, SPRY1, C2GNT, Serpine1
  • lower expression in bright cells
    • Galectin-1, OSTF epsilon, TGFBR3, TP epsilon, CD6, SLC16A6, TNFR-II, GZM A (Granzyme A), KIR3DL2, KIR3DL1, KIR2DL3, RORA, GZM B, SCYA5, MIP-1A, MIP-1B, CD16 (22 fold)

Interestingly gene expression of CD56 is nearly identical although flow cytometry demonstrates a difference in protein level of 5 to 10 fold. CD56 is slightly higher expressed in mRNA of CD56bright cells.

Minimal differences in gene expression were identified for the following genes:

  • CD63, CD56, NKG2C, CD59, NKG2E, DEGS, TSSC3
  • RAC2, KIR2DL4, SLC9A6, DAD1, DEFCAP, HCS, DAG1, MPV17, CD53, NK4, FASL, CTNNA1, PRF 1 (Perforin)

Dzionek, Turkiewicz, Lokan et al. (2005) compared gene expression of MACS selected NK cell subsets using PIQOR immunology arrays. 11 genes (out of 1076 present on the array) were significantly differently expressed:

  • RAR, TNFSF6, AIOLOS, MCP, CD9, CDH3, CCL3-CCl3L1, CCL5, NOTCH2, PECAM1, PRKCZ
